राम पोथीनेनी शुरू से ही एक्टर बनना चाहते थे. साउथ फिल्म इंडस्ट्री में उनके एक्टिंग करियर की शुरुवात साल 2002 में आई एक शॉर्ट फिल्म Adayaalam से होती है। इस फिल्म में वे एक 18 साल के ड्रग एडिक्ट लड़के का किरदार निभत्ते हैं. अगर फिल्मों में उनके डेब्यु की बात की जाए तो उन्होंने साल 2006 में फिल्म Devadasu (Ram Pothineni Debut Movie) से डेब्यु किया था. इसके बाद राम ने फिल्म Jagadam की जो काफी ज्यादा हिट रही. इस हिट फिल्म के बाद राम का करियर चल निकला और उन्होंने कई हिट फिल्में की.
